چکیده

Cytochrome P450 (P450) enzymes primarily catalyze mixed-function oxidation reactions, plus some reductions and rearrangements of oxygenated species, e.g. prostaglandins. Most of these reactions can be rationalized in a paradigm involving Compound I, a high-valent iron-oxygen complex (FeO), to explain seemingly unusual reactions including ring couplings, ring expansion and contraction, and fusion of substrates. Most P450s interact with flavoenzymes or iron-sulfur proteins to receive electrons from NAD(P)H. In some cases P450s are fused to protein partners. Other P450s catalyze non-redox isomerization reactions. A number of permutations on the P450 theme reveal the diversity of cytochrome P450 form and function.

Advances in mechanisms of activation and deactivation of environmental chemicals.

Environmental chemicals are both activated and detoxified by phase I and phase II enzymes. The principal enzymes involved in phase I reactions are the cytochrome P-450s. The phase II enzymes include hydrolase and the conjugative enzymes such as glucuronyltransferases, glutathione transferases, N-acetyltransferase, and sulfotransferase. Although other phase I and phase II enzymes exist, the pres...

Reactions and significance of cytochrome P-450 enzymes.

Cytochrome P-450 enzymes (EC 1.14.14.1, nonspecific monooxygenase) catalyze the oxidations of many chemicals (1). The mass of the substrates ranges from that of ethylene (Mr 28) to that of cyclosporin A ( M , 1201). The classification of a hemoprotein as a P-450’ is defined by its absorption spectrum; the Fe(I1)CO complex has a characteristic absorption maximum (Soret band) near 450 nm due to a...

Cytochrome P-450 isozymes and monooxygenase activity in aquatic animals.

The roles of different forms of cytochrome P-450 in activation and deactivation of toxic chemicals, synthesis and breakdown of steroid hormones, and other functions, indicate the significance of these enzymes. Monooxygenase systems have been studied in species from several phyla of aquatic organisms. However, cytochrome P-450, the dominant catalyst in xenobiotic monooxygenase activity, is best ...

Comparison of cytochrome P-450- and peroxidase-mediated activations of carcinogenic azo dyes and N-nitrosamines.

Carcinogenic azo dyes (dimethylaminoazobenzene, Sudan I) and N-nitrosamines (N-nitrosomethylaniline, N-nitrosomethylbenzylamine) are oxidized by cytochrome P-450 isoenzymes and peroxidase yielding metabolites which in vitro bind to DNA and transfer RNA (tRNA). The parallelism and differences in oxidative reactions are described. Peroxidase is more effective than P-450 in activating reactions of...

Oxygenated cytochrome P-450cam: evidence against axial histidine ligation of iron.

Because of their unusual spectral and catalytic properties, the cytochrome P-4.50 enzymes have been intensely investigated [ 11. A major objective has been to identify the non-porphyrin ligand(s) to the central iron in states l-5 of the P-450 reaction cycle (see Scheme 1). Comparative physical properties of synthetic porphyrin complexes with various axial ligands, myoglobin complexes with axial...
